To determine how single nucleotide polymorphisms (SNPs) in the hypoxia inducible factor-1α (HIF-1α) gene coding regions affect gastric cancer, the authors conducted an association study of the HIF-1α polymorphisms C1772T and G1790A for a Tibet population. DNA was extracted from peripheral blood of 87 gastric cancer patients and 106 controls and analyzed using the polymerase chain reaction/ligase detection reaction test for HIF-1α polymorphisms. There was a significant increase in the frequency of the GA 1790 genotype in patients with gastric cancer compared with healthy controls (OR 2.93; 95% CI 1.06–8.06). The genotype frequency of the HIF-1α G1790A allele A is higher in gastric cancer groups than in controls (OR 2.78; 95% CI 1.03–7.45). As for the C1772T polymorphism, no positive correlation was found between gastric cancer patients and controls (P = 0.06). Our results suggest that the HIF-1α G1790A polymorphism may be associated with gastric cancer in Tibetans. 相似文献

The aim of this study was to investigate the role of miR-138 in osteoporosis and its underlying mechanism. Hydrogen peroxide (H2O2) was used to induce osteoporotic injury of osteoblasts. The cell viability and apoptosis of MC3T3-E1 cells was assessed using MTT assay and flow cytometry, respectively. The cell transfection was carried out to modulate the expression levels of miR-138 and TIMP-1 in MC3T3-E1 cells. Luciferase reporter gene assay was performed to determine the interaction between miR-138 and TIMP-1 3′UTR. In the present study, H2O2 inhibited osteoblasts growth and induced intracellular endoplasmic reticulum (ER) stress accompanied by high expression of miR-138. We also confirmed that miR-138 promoted osteoblasts apoptosis in vitro and in vivo. MiR-138 was further indicated to inhibit osteoblast survival via negative regulating TIMP-1 expression. Moreover, the downregulated TIMP-1 also mediated the ER stress-induced apoptosis of osteoblasts. We confirmed that miR-138 and ER stress were induced in osteoporosis and then promoted the apoptosis of osteoblasts, at least in part, through TIMP-1. 相似文献

Plasmon‐enhanced gold nanorod (AuNR) with high photothermal conversion efficiency is a promising light‐controllable nanodrug delivery system for cancer therapy. Understanding the mechanism for the light‐controllable drug release of AuNR delivery systems is important for the development of nanomedicine. In this study, the rhodamine B (RB) released from AuNR‐RB nanodelivery system was quantitated and visualized by using two‐photon luminescence (TPL) imaging combined with correlation spectroscopy. The photofragmentation of AuNR induced by femtosecond pulsed laser was revealed by TPL correlation spectroscopy when the laser energy was above the thermal damage threshold of AuNR, and the RB released from this nanodrug delivery system was visualized by TPL imaging. Furthermore, the photofragmentation‐induced release of RB from AuNR‐RB nanodelivery system was visualized in living MCF‐7 breast cancer cells by TPL imaging combined with correlation spectroscopy. These results provided a novel optical approach to quantify the release of drugs from gold nanocarriers in complex biological media. 相似文献
